A DOT specimen collection is the process of collecting a urine specimen for federally regulated drug testing in accordance with the U.S. Department of Transportation (DOT) regulations (49 CFR Part 40). The collection is performed using strict procedures designed to protect the integrity of the testing process.

DOT drug testing applies to employees and applicants who perform safety-sensitive duties for employers regulated by agencies such as the Federal Motor Carrier Safety Administration (FMCSA), Federal Aviation Administration (FAA), Federal Railroad Administration (FRA), Federal Transit Administration (FTA), Pipeline and Hazardous Materials Safety Administration (PHMSA), and the United States Coast Guard (USCG).
